CJC-1295 vs Sermorelin: Structure, Duration, Evidence and Approval History

Compare two GHRH receptor agonists by molecular design, duration, human evidence, historical approval and current regulatory status.

Executive Summary

CJC-1295 and Sermorelin both activate the GHRH receptor, but they differ in molecular design, exposure and regulatory history. Sermorelin is the native active GHRH (1-29) fragment and had historically approved U.S. products. CJC-1295 contains stabilizing changes and, in its DAC form, binds albumin for multi-day exposure. Neither is currently an FDA-approved general anti-aging, bodybuilding or weight-loss therapy.

Shared Pathway

Both compounds act upstream of pituitary GH release through the GHRH receptor. Increased GH can raise hepatic IGF-1. This shared mechanism explains why both appear in the same category but does not make their products or evidence interchangeable.

Structural Difference

Sermorelin reproduces the first 29 amino acids of GHRH. CJC-1295 modifies residues to resist enzymatic breakdown, and the DAC version adds an albumin-binding group. These changes create a much longer exposure profile.

Human Evidence

Sermorelin has a longer history in diagnostic and pediatric endocrine use. CJC-1295 has controlled healthy-adult pharmacokinetic evidence demonstrating sustained GH and IGF-1 increases. Neither evidence base establishes broad healthy-adult anti-aging or performance outcomes.

Approval Status

Sermorelin products were historically approved but later discontinued. A current compounded Sermorelin preparation is not itself an FDA-approved product. CJC-1295 has never been FDA approved. Approval history must be described at the product and indication level.

Key Insight

CJC-1295 offers prolonged exposure; Sermorelin offers longer clinical history. Those facts do not create a universal clinical winner.
